Notes2 g of chocolate was weighed into a 50 mL centrifuge tube. The sample was brought to a total volume of 40 mL with IPA. The sample was then sonicated at 40 °C for 5 minutes followed by gentle mixing by hand. This was repeated for an additional two cycles. The sample was stored in a -20 °C freezer for 30 minutes to allow the lipids to precipitate. The sample was then vortexed for 5 seconds followed by centrifugation at 3000 rpm for 5 minutes. The supernatant was diluted 10-fold in 25:75 water:methanol, vortexed briefly, and filtered using a 0.2 μm Thomson SINGLE StEP standard filter vial (cat.#: 25893) prior to analysis.
